Supplemental movies for Molecular and Cellular Biology

Supplementary movies 1, 2 and 3. The videos illustrate the phenomena depicted by figure 1. Movie 1 demonstrates EB3-GFP, "comet tails" in the axon. The movie contains 29 images, taken at intervals of 12 seconds. Movie 2 demonstrates the non-polar orientation of microtubules in the soma and the typical MTOC usually observed at the axon hillock as determined by EB3-GFP.  The movie is composed of 70 images, taken at intervals of 4 seconds. Movie 3 illustrates the transport of SR101 labeled endocytotic vesicles. The movie comprises 20 images, taken at intervals of 7.2 seconds. The frames are shown at a rate of 10/s. Scalebars: 10 Ám.

Supplementary movies 4, 5 and 6. MTOCLS and aberrant axoplasmic transport. The videos present the MTOCLS shown in figure 3.  Movie 4 (Fig 3 A ) a MTOCLS taken 24h post tau microinjection. The movie comprises 30 images, taken at intervals of 4.4 seconds. The frames are shown at a rate of 10/s.

 Movie 5 presents the MTOCLS of figure 3D. in green EB3-GFP. Movie 6 acquired from the neuron shown in 3F reveals jamming of SR101 labeled vesicles. Movies 5 and 6 are composed of 30 images, taken at intervals of 5.5 seconds. The frames are shown at a rate of 10/s.  Scalebars: 10 Ám.

Supplementary movies 7 and 8. Loss of microtubules uniform polarity is accompanied by aberrant transport.  The movies relate to Figure 4. Movie 7 depicts a tau expressing neuron wherein EB3-GFP comet tails point in various directions. SR101 labeled vesicles are presented in movie 8 and were predominately stationary. The movies comprise 30 images, taken at intervals of 5.3 seconds. The frames are shown at a rate of 10/s.  Scalebars: 10 Ám.

supplementary movies 9 and 10. Accumulation of EB3 at the submembranal domains.  The movies relate to Figure 5.  Movie 9 illustrates a B neuron  characterized by accumulation of tau at the submembranal domains. The EB3-GFP comet tails reside mainly along the axonal cortex, some EB3-GFP comet tails at the axonal core point their plus ends in various directions. Movie 10 portrays endocytotic retrogradely transported vesicles labeled by SR101. While the transport of SR101 labeled vesicles came to a halt within the axonal core, some transport is retained at the axonal cortex. Movies comprise 45 images, taken at intervals of 4.6 seconds. The frames are shown at a rate of 10/s.  Scalebars: 10 Ám.
